I've just relocated my battery to the boot of my Holden Torana LC.
Now I have a Battery that is sealed with battery box fitted. I also have the vent tubes.
Now the battery has 2 vent ports and both are opposite sides of the battery.

My question is... Do I have to place hoses in both vent ports or can I just block one so it only goes out one side only?

or is it the way it works as one vent is for 1 set of cells to vent while the 2nd vent does the other set of cells?

The left hole is a Vent Port. and on the other side of the battery is the other one.
(The Right hole is nothing -basically used for the handle hinge)

It's the only thing left to do then I can finally fit Triple SU's in as battery was in the way

Theoretically you can safely block one end and vent it but in the instance of a prjoect car that might see the battery flatten.more than normal, id tie them both into a Y piece and vent into a larger hose directly out the boot allowing a vacuum effect to draw the gases out.

I fitted hoses to both sides and rerouted them to a 'Tee' Piece and drilled a hole into the side of the battery box. Hose exits the hole and lead it to outside the vehicle.

Cheers. Looks really professional as it's not in the way of the Battery Positive and Negative wires.
