I have a 2000 VT II Berlina that's approaching 250 thousand kms. Is this going to be a major service? Also, any faults I should start looking out for?

Well When was your last major service ?
A Major service is every 40,000kms.
Requires.
Leads, Plugs, Air/Fuel/Oil Filter, Oil Change at least.
Also depending how long ago.
Transmission service, Brake fluid change, Coolant change & Flush, Power steering Flush.
& Checked all your gaskets, look for leaks of any sorts. Oil Locks & Door hinges, Tyre pressures/ Tread where. Lights, Coolant hoses, Brake pad/ disc wear, Suspension, signs of rust.
That should give you a good starting point.
Last major service was a 200,000, so I'm thinking it's probably overdue.
Also, it's been running a bit dodgy lately, spluttering at times, rough idle and it's cut out when cold a few times, both in drive and reverse. Could that be spark plugs, does it need a tuneup or something worse?
